聚乙烯亞胺(細(xì)胞培養(yǎng))(Polyethyleneimine;PEI;Epomine)
將外源基因?qū)胝婧思?xì)胞是分子生物學(xué)研究中的一項(xiàng)重要技術(shù)。轉(zhuǎn)基因載體主要分為病毒載體系統(tǒng)和非病毒載體系統(tǒng)。病毒載體系統(tǒng)轉(zhuǎn)染效率高,是體內(nèi)基因治療的主要工具,但安全性存在隱患,且有免疫原性,體內(nèi)不能反復(fù)應(yīng)用,制備較復(fù)雜,而且所能裝載的外源DNA大小有限,使其應(yīng)用受到很大限制。非病毒載體是病毒載體的重要補(bǔ)充途徑,一般認(rèn)為安全性高,載體一般無免疫原性,而且是基于質(zhì)粒的轉(zhuǎn)染,易于組裝[1-2]。目前已有的非病毒型載體介導(dǎo)基因轉(zhuǎn)染的方法有裸DNA注射、磷酸鈣介導(dǎo)、電轉(zhuǎn)移法、陽離子脂質(zhì)體及陽離子聚合物介導(dǎo)等。1995年,Boussif首次報(bào)道了聚乙烯亞胺(細(xì)胞培養(yǎng))(polyethylenimine,PEI)可作為非病毒載體,其后,聚乙烯亞胺成為陽離子多聚物研究zui快的領(lǐng)域。為了研究PEI基因轉(zhuǎn)染試劑的轉(zhuǎn)染效果,本文通過聚乙烯亞胺(細(xì)胞培養(yǎng))PEI轉(zhuǎn)染表達(dá)綠色熒光蛋白的pEGFP質(zhì)粒到293T細(xì)胞,以觀察其效果,并測定了多種因素對(duì)轉(zhuǎn)染效率的影響。
