 血管內(nèi)皮細(xì)胞對維持血管動態(tài)平衡起著重要作用。它們合成、分泌凝血和纖溶系統(tǒng)的激活因子和抑制因子、影響血小板粘附和聚集的調(diào)節(jié)因子。內(nèi)皮細(xì)胞還釋放控制細(xì)胞增殖和調(diào)節(jié)血管壁緊張度的分子。這些過程都可以用培養(yǎng)的細(xì)胞進(jìn)行體外研究，人臍靜脈和動脈內(nèi)皮細(xì)胞是這類研究中zui常用的細(xì)胞類型。除了普通的內(nèi)皮細(xì)胞特征例如“鵝卵石狀”，VIII因子陽性著色，能攝取乙?；兔芏戎鞍祝四氀軆?nèi)皮細(xì)CD31染色陽性。用IL-1或TNF-α預(yù)先處理過的細(xì)胞也能選擇性地表達(dá)E-selectin和VCAM。  人臍靜脈內(nèi)皮細(xì)胞(HUVEC)提取于人臍帶靜脈組織，原代凍存。每管含有細(xì)胞數(shù) 5×105 cells/ml，此細(xì)胞通過vWF/Factor VIII 和CD31 (P-CAM)免疫熒光染色驗證，經(jīng)測試不含有HIV-1、HBV、HCV、支原體、細(xì)菌、酵母和真菌。細(xì)胞可以達(dá)到15倍增。

The vascular endothelial cells contribute to the maintenance of vascular homeostasis. They synthesize and secrete activators as well as inhibitors of both the coagulation system and the fibrinolysis system in addition to mediators that influence the adhesion and aggregation of blood plaets. Endothelial cells also release molecules that control cell proliferation and modulate vessel wall tone. Many of the endothelial processes can be studied in vitro using cultured cells, and human umbilical vein endothelial cells (HUVEC) are the most commonly used cell type for such studies. Except for the common endothelial cell features such as "cobblestone" morphology, positive staining for Factors VIII and the ability to take up acetylated low-density lipoprotein [1], HUVEC stein positively with CD-31 [2]. Cells pretreated with IL-1 or TNF-alpha also selectively express E-selectin [3] and VCAM [4].
HUVEC from ScienCell Research Laboratories are isolated from human umbilical vein. HUVEC are cryopreserved at passage one and delivered frozen. Each vial contains 5 x 105 cells in 1 ml volume. HUVEC are characterized by immunofluorescent method with antibodies to vWF/Factor VIII and CD31 (P-CAM) and by uptake of DiI-Ac-LDL. HUVEC are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast and fungi. HUVEC are guaranteed to further expand for 15 population doublings at the conditions provided by ScienCell Research Laboratories.
